I would like to build a drone that can fly for 30-45 min with a payload of 100 lbs.

That sounds like a heavy-duty drone haha. I suggest you take a look at the How to Make a Drone / UAV tutorial series, it will guide you through the process of building your own drone and you will also find some useful links for the parts you are going to need.
