Is it possible to connect them directly? Is the port extender able to switch the relays? Are the 25mA per port sufficient?

Unfortunately, these 2 modules aren’t plug and play, you will need to wire your setup manually with jumper or wires.

From what I could read, the coil is drawing around 0,07A at 5V.
